yes, you need only one glyph of each type to cast all the spells that glyph can be a part of.

Well is it possible to use the crystal arrow and darkness weakness glyphs to form an arrow of weakening?

Combinations and spells are predetermined - you won't be able to mix just anything you want. You'll have to unlock certain combinations for certain spells.

you also need to be high enough in that certain way of magic or you won't discover and combination as well.

so if you know that two glyphs make a spell, but you are not high enough to cast it, you won't discover the spell even if you do have the right combination, that is, until you train enough to be able to cast it, and then you could discover it.
